主人公邁克·安瑟林（約翰·庫薩克飾）是一個恐怖小說家。將裝神弄鬼作為本職工作的邁克，平日里卻是個徹頭徹尾的無神論者。為了完成自己的新書，邁克決定找一家「鬧鬼」的房間住上十天，而膽大包天的他最終選擇了傳聞中最陰森恐怖的海豚酒店的1408號房間。

酒店經理傑拉爾德·奧林（塞繆爾·傑克遜飾）拿出在那個房間中歷年來數十起離奇死亡事件的照片，來力勸邁克不要冒險，但邁克滿不在乎，堅持著自己的選擇。而在搬進1408號房間後，一件件離奇的超自然現象讓邁克徹底改變了自己的鬼神人生觀。大量恐怖驚悚的真實體驗為邁克的新書著實積累了素材，但問題的關鍵是，他必須得先活著走出那個房間。
